Another thought 💭 25mg avg attacks: 6 (1 high patient) 50mg: 1.15 (no high) 75mg: 7.7 (1 high/1 mid) Curious if ⬆️ attack rates correlate to ⬆️ protein levels.. you’d assume yes. Positive that 50mg had lower attack rate baseline — more representative of overall #HAE pop

$NTLA-2002 50mg cohort math challenge 😅 74, 79, 89, 94 These are possible numbers I got that average 84 and have sample standard deviation of ~9 Seems highly likely that the lowest was above 70% protein reduction. Much less variability than 25mg cohort. 25mg had SD= ~26

“High level” math by me and @JinghangHu because IR departments seem to never reply 🤷🏻‍♂️ 50mg cohort was the key I wanted to figure. SD of 9, Mean of 84 means we have low variability! 78 82 88 89 Could be a very close estimate to the actual protein level reductions at 50mg!!!

These type of charts by Intellia really annoy me… are you really going to use standard deviation when your sample sizes are 3, 4, and 3 🤦🏻‍♂️ I’d like to think we could simply put each patient’s percentage down! Not to mention how the lines jumble all together.

I’m hoping all 4 in the 50mg dose were between ~74-94% (avg 84). This would give us greater confidence that the 50 won’t have such drastic variability moving forward. I want $NTLA to be able to say “cure” & I know 75mg would’ve provided that. 50mg could if much lower variability

One analyst was smart and got them to tell us that the one who didn’t reach 60% only had 40% reduction in the 25mg cohort. You’d think having 1/3 patients not reach your 60% threshold would automatically throw out that dose level. I have sent an email to IR @intelliatx

They did not break down the 4 patients in the 50mg cohort but I’d assume you’ll see 1/3 of patients in each area: 70%, 80%, 90%. But the 25mg variability is scary to me. One only had 40% so other 2 averaged 80.5%. HUGE variability @ 25mg makes decent variability possible at 50mg

The one 25mg patient who had one attack after the 16 week period due to a sports injury only had 40% knockdown! 60% is thought to be the threshold for success. 67% avg for 25mg (n=3) 84% for 50mg (n=4) 95% for 75mg (n=3)

🧵 It makes literally zero sense to me that $NTLA would take forward the 25mg (in addition to 50mg) dose. There is literally zero chance that ends up as the dose. Would’ve been much smarter to bring forward the 75mg to go with the 50mg. 🧵👇

$45.80 was the $NTLA offering price from Nov 2022. I don’t believe we’ve closed over that ever since. Has been a battleground level I’d love to close over! Presentation is now up for those who missed it live early this morning. Will watch now.
